Abstract: Sweetpotato (Ipomoea batatas (L.) Lam.) is a globally important crop for food, feed, industrial raw materials, as well as functional foods. With the rapid development of biotechnology such as genomics, transcriptomics, metabolomics, and gene editing, significant progress has been made in exploring genes related to important traits in sweetpotato. The current status is systematically reviewed in mining excellent genes of seven key trait areas—yield and yield components, quality, biotic and abiotic stress resistance, resource use efficiency, suitability for mechanization, and storage properties—both domestically and internationally since the 14th five-year plan. It focuses on summarizing the functions and molecular mechanisms of key genes controlling storage root formation and bulking, starch and sugar metabolism, anthocyanin and carotenoid biosynthesis, disease and pest resistance, drought and salt tolerance, and plant architecture regulation. The core challenges of molecular biology research of sweetpotato were summarized. Finally, prospects for future molecular design breeding in sweetpotato are outlined. It is proposed to strengthen the integration of multi-omics and the application of gene editing technologies, construct high-precision genetic maps, deeply explore and utilize wild germplasm resources, and establish efficient breeding systems for pyramiding multiple traits. These efforts aim to accelerate the development of breakthrough sweetpotato varieties with high yield, superior quality, multi-resistance, suitability for mechanization, and high resource use efficiency, thereby supporting the industry for sustainable development.
